The purpose of the UCLA Molecular Genetic Technology Center is to provide efficient, cost effective services to the UCLA research community. Dr. Hong Wu became the director of the Center on 4/1/2003 and the Center has been administrated through the Department of Molecular and Medical Pharmacology since. The Center consists of three service cores: the ES cell core, the injection core, and the technology development core. During FY03, we provided 144 services to 35 research laboratories, including 4 labs outside of UCLA.

The ES cell core is located in the Department of Molecular Pharmacology at the second floor of CHS building. Under the directorship of Dr. Xin Liu, the ES cell core provides services, including: 1) consultation for designing transgenic and knock-out constructs; 2) providing basic vectors for various knock-out and knock-in constructs; 3) electroporation of ES cells and providing genomic DNA harvested from up to 300 ES clones; 4) expanding ES clones either generated by the core or by outside facility; and 5) custom deriving ES cell lines from animals.

Dr. Meisheng Jiang, an Assistant Professor of Pharmacology, became the director of the injection core on 4/1/2003. The injection core provides services such as 1) ES cell injection and generation of chimeras; 2) generating transgenic founders; and 3) providing inbred strains for back-crossing of chimeric mice. The injection core is located in the Gonda Variorum.

To adapt and improve the current technologies to meet the future needs for molecular genetic studies, a technology development core has been added to the Center. Headed by Drs. Xin Liu and Hong Wu, the core is temporarily located in CHS 2V-111. The targets of this core for the next two years include: 1) providing germline tested C57 ES cells for gene targeting; and 2) providing tetraploid injection service, a new technology which will shorten the time required for generating a knock-out mouse by at least 6 months.
